The Molecular Identification of Hypericum Perforatum L. Growing in Nineveh Governorate

This research focused on studying the molecular genetic marker of the wild St. John's Wort plant (Hypericum perforatum L.), which was collected in September from Nineveh Governorate (Tal Afar District / Zummar Subdistrict). The studied samples were taken from newly grown leaf tissues aged 3–4 weeks, free from insect and disease infestations. The analysis utilized the Specific-PCR technique, where DNA fragments were amplified using the polymerase chain reaction (PCR) method and the Favor PrepTM Plant Genomic DNA Extraction Mini Kit. The results of this study matched the nucleotide sequences recorded in the National Center for Biotechnology Information (NCBI) GenBank database for the plant species under investigation. This was achieved by using the primer pair (ITS1F: 5'-TCCGTAGGTGAACCTGCGG-3') and (ITS4R:5'-TCCTCCGCTTATTGATAT GC-3').
After inputting the nucleotide sequences into the NCBI-BLAST tool and the MEGA 6.0 software, the analysis demonstrated a high degree of similarity with the registered genes in the GenBank database.
